HEPES is a non-ionic amphoteric buffer with a pH range of 6.8 - 8.2 and a good capacity in the pH range of 7.2 - 7.4. It is widely used in cell culture, IVD detection kits, DNA/RNA extraction kits, PCR diagnostic kits, biomedicine and other fields.
HEPES buffer has no toxic effect on cells, and can control a constant pH range for a long time. It is often used in the buffer system of cell culture. The common working concentration is 10-25mM. Generally, the medium containing 20mmol/L of HEPES can achieve the buffer capacity. . Compared with bicarbonate buffer, HEPES showed better pH control ability in tissue or organ culture. It is also used as a buffer for quantitative and selective determination of antigen-antibody reactions.
One of the applications of HEPES is Onivyde. In 2015, Onivyde was approved by the U.S. Food and Drug Administration (FDA) for new drug marketing. In October 2016, Onivyde received new drug marketing approval from the European Medicines Agency (EMA). In April 2022, Onivyde was officially approved by China's National Medical Products Administration (NMPA) for the treatment of metastatic pancreatic cancer.
