1. Liposomes are composed of phospholipids as the skeleton membrane and additives. The phospholipids used to prepare liposomes include natural phospholipids and synthetic phospholipids.

2. Additives, such as cholesterol, are needed to prepare liposomes. liposome phospholipid undergoes a mixture with  Cholesterol, which is  also an amphiphilic substancear, to prepare stable liposomes. Its function is to adjust the fluidity of the bilayer and reduce the liposomes membrane permeability.

3. In the liposome formulation, special excipients are also utilized. For example, irinotecan liposome Onivyde® uses sucrose octasulfate for gradient drug loading.

4. The lyophilized liposome generally uses sucrose or trehalose as the cryoprotectant for lyophilization.
