SM-102 carries a positive charge and exhibits excellent biocompatibility, which enables it to form complexes with negatively charged nucleic acids, to protect nucleic acids from degradation and facilitate their entry into cells and subsequent release into the cytoplasm.
Using LNPs formulated with SM-102 to deliver luciferase mRNA can induce luciferase expression in the mouse liver, demonstrating the significance of SM-102 in drug delivery and gene transfection.
DLin-MC3-DMA is a colorless to yellowish oily liquid with a pKa of 6.44. The functional activity of LNPs is correlated with the pKa value of the cationic lipid used. LNPs formed with DLin-MC3-DMA with a pKa of 6.2 to 6.5 exhibit higher gene-silencing activity.
