DOTAP is an effective cationic lipid, originally utilized in commercial products as a gene transfection reagent. It is also applicable as a pharmaceutical excipient in liposomal formulations. Its notable application is EndoTAG-1, a cationic liposomal formulation of paclitaxel. DOTAP imparts a positive charge to EndoTAG-1, allowing it to selectively adhere to tumor endothelial cells (TECs) with negative charges. This selective adsorption facilitates targeted attack on tumor neovasculature without harming healthy vessels, effectively inhibiting tumor growth.

Targeted Delivery: DOTAP can serve as a supplementary component in selective organ targeting (SORT) lipid nanoparticles (LNPs). Incorporating 10%-15% DOTAP in LNPs facilitates targeted drug delivery to the spleen, while increasing the DOTAP content to 50% achieves optimal lung delivery efficiency.
DOTAP is biologically inert, biodegradable, non-toxic, and non-immunogenic. Its polar head stably binds to negatively charged DNA molecules through electrostatic interactions, forming stable complexes spontaneously that enter cells and deliver nucleic acids into the cytoplasm without causing damage to the organism. It is therefore often used as a vector for eukaryotic transfection reagents and DNA vaccines.
